Infrastructural Centre Planta (IC Planta)

The Infrastructural Centre Planta (IC Planta) is one of three program and organization integrated infrastructural centres that forms the Infrastructural Program of the National Institute of Biology (IP NIB).

IC Planta’s equipment supports research activities, bodies of ministries, enterprises and educational activities. IC Planta ensures collaboration between researchers of different research programs, projects and institutions. It facilitates connections of researchers with the users of this research, which are other budget users and various industries, as well as facilitating connections between research activities and educational processes.



IC Planta has existed since 1991, first as the Technological Core – Centre for tissue culture and virology. The Technological Centre Planta was founded in 1994 by the National Institute of Biology, the pharmaceutical company Krka d.d. and the seed company Semenarna d.o.o. In 1998 the Technological Centre Planta was retransformed to the Instrumental Centre Planta. Since then it has been very strongly connected to the Chair of Zoology at the Department of Biology at Biotechnical Faculty of the University of Ljubljana, led by Prof. Dr. Jasna Štrus in the segment of electron microscopy. Since 2004, IC Planta operates as an infrastructural centre.

All large equipment of IC Planta is technologically advanced. High technological equipment demands careful, regular and professional maintenance. Each piece of equipment has its caretaker and his substitutes that take care of regular maintenance and impeccable functioning of the equipment. IC Planta’s equipment is managed and kept by highly skilled workers as we facilitate adequate permanent education of the employees. IC Planta has modern and well treated research equipment (in accordance with ISO/IEC 17025 system) which is an essential condition for competitive research performance in life sciences. Quality and well maintained auxiliary equipment are a must for a proper functioning of the large infrastructural equipment. Thus the auxiliary equipment is subjected to special care and ongoing modernization.

IC Planta purchased large infrastructural equipment by merging funds from different sources and institutions. The large equipment is also used by small research programs, research projects and other users that cannot afford purchasing and keeping such expensive equipment. Such a broad spectrum of use assures maximal exploitation of the equipment and consequently a good yield of all invested funds. Training courses are organised for frequent users, and services regarding the use of the large equipment of the IC Planta are offered to infrequent users.

Before purchasing any new equipment, IC Planta studies very carefully whether a certain device is already available in Slovenia in order to avoid the unreasonable duplicating of equipmentand to assure reasonable complementation of equipment. With its equipment IC Planta joins laboratory networks, which enables its users to use the equipment of different infrastructural programs in different stages of research and applications, where IC Planta’s equipment plays an important role. In the field of molecular biology IC Planta is linked to other centres through the Department of Biotechnology and Systems Biology,: the Centre for Functional Genomics and Bio-Chips with the main offices at the Medical Faculty of the University of Ljubljana, the Infrastructural Centre for Surface Plasmon Resonance with the main offices at the Biotechnical Faculty of the University of Ljubljana and the Centre for Protein Production and Structure located at Jožef Stefan Institute. Besides, IC Planta takes an active role in the Slovenian technological network Plants for the Future.

IC Planta is part of the Department of Biotechnology and Systems Biology at the National Institute of Biology (NIB).

The large infrastructural equipment of IC Planta consists of:



Transmission electron microscope (Philips CM100) with CCD cameras Orius SC200 (Gatan) in BioScan 792 (Gatan)
Transmission electron microscope (TEM) (Philips CM100) with two digital cameras BioScan 792 (Gatan) and Orius SC200 (Gatan), and with software DigitalMicrograph for image analyses.

TEM operates on the same basic principles as the light microscope but uses electrons instead of light. Cathode is source of electrons which are accelerated by an anode and focused by electromagnetic lenses into a very thin beam. The electron beam travels through a specimen. Image is function of electrons scattering on the elements in the specimen. Image can be observed either on a fluorescent screen, or captured by CCD camera in real time and stored in digital format. CCD camera Orius SC200 (Gatan) has extremely high resolution (4M pixles) and therefore enables observation of single macromoleculs and precise analyses of structures (e.g. surface of viruses). CCD camera BioScan 792 (Gatan) has lower resolution, but extremely wide field of view. Therefore the TEM with both cameras enables high practicability that with only one camera would not be possible.

TEM Philips CM 100 is successfully used in basic and applied biological studies. The resolution makes it possible to observe samples on the ultrastructure level. Regarding biological samples two methods are used for sample preparation. The negative staining method is suitable for observing particles in a solution e. g. viruses, bacteria, macromolecules (proteins, nucleic acids, liposomes). A second method enables visualization of plant, animal and human tissues on the nanometre scale. For cell ultrastructure visualization samples are fixed, embedded and cut into thin slices. TEM Philips CM 100 is used for analyses of structure and immunolocalization in different biological samples.

Transmission electron microscope Talos L120C
Transmission electron microscope (TEM) (Talos L120C) is specialized for the observations of biological specimens. Its characteristics are optimally adjusted for the observations of biological specimens. It has high contrast and high resolution at a wide magnification range, it enables fast scanning of a specimen on screen and capturing and processing of high resolution images. These characteristics make it possible to visualize a very wide range of biological samples, ranging from isolated macromolecules, viruses and bacteria to the ultracellular structure of fungi, plants, animals and humans.

TEM Talos L120C is co-owned by the National Institute of Biology (NIB), Department of Biology at the Biotechnical Faculty of the University of Ljubljana and National institute of chemistry (KI).

System for sample preparation for transmission electron microscopy
System for sample preparation for transmission electron microscopy (TEM) includes equipment for preparation of small particles, like viruses, proteins and liposomes with negative staining method as well as preparation of plant and animal tissues and cell cultures for ultrastructure observation. Instrument for preparation of grids (LeicaEICA EM ACE200) is used for preparation of grids for negative staining method and has carbon thread coater for evaporation of carbon of definite thickness and glow discharge unit. Glassknifemaker (LeicaEICA EM KMR3) enables preparation of glass knives from glass strips. System includes also Hotplate for mounting of trufs on glass knives. Glass knives are used for preparation of semi-thin slices of embedding biological samples using ultracut (LeicaEICA ULTRACUT UC7). Besides semi-thin slices ultracut is used also for preparation of ultra-thin slices. Ultracut UC7 has multi-LED-illumination for standard and back light illumination, specimen transillumination and spot light illumination. Touch-sensitive-control unit enables to determine cutting speed from 0.05 to 100 mm/s, cutting window, step control of movement, 3 selectable return speed and other functions. Light microscope (LeicaEICA DM 1000 LED) used for observation of semi-thin slices is equipped with digital camera FLEXAM C1 with CMOS sensor. Inverted confocal laser scanning microscope (Leica Stellaris 5)
Inverted confocal laser scanning microscope (Leica Stellaris 5) is a new generation inverted confocal microscope that is used to observe the locations of molecules and the interactions between molecules in living plants in real time without prior preparation. It has an ultra-sensitive HyD S detector that enables faster image capture, which is key to monitoring internal changes in real time. In addition, the new detector provides a better signal-to-noise ratio and signal accumulation. It allows the selection of an arbitrary range of light wavelengths, in this way there is no disturbing background in the images, which is a result of the non-specific fluorescence of the investigated sample, and it is also possible to monitor several labeled molecules in the cell at the same time. Confocal stereomicroscope (Leica TCS LSI)
Confocal stereomicroscope (Leica TCS LSI) enables non-invasive observation of the fluorescence in the samples. The speciality of the Confocal stereomicroscope (Leica TCS LSI) is broad spectrum of magnifications from 0.7x (16 mm field of view) to 20x. Therefore larger object as well as single cells could be investigated. Stereomicroscope has long working distance what enables investigation of big objects (e.g. plants in pots). One of the most important advantages of confocal stereomicroscope over the relative techniques is the possibility of the observation of intact tissues and organs what shortens the time of the analyses and significantly increases the relevance of the results.

Whole plant imaging system Newton 7.0. BIO (Vilber)
Whole plant imaging system Newton 7.0. BIO (Vilber) enables following bioluminescence and fluorescence in intact plants in spatial and temporal manner. Dimensions of the chamber and field of vision enable imaging of samples of different sizes, from seedlings to plants of medium height (approx. 35 cm high and 20 cm wide). Highly sensitive camera with aperture of f/0,7 and low read noise enable high dynamic range and spatial resolution of images. Wide spectral range of the camera (400-900 nm) and a set of 8 filters enable detection and separation of the signal of multiple fluorescent proteins. The system for multi-level phenotypic characterization of plants with microorganisms includes a third module, namely the system for molecular characterization of nucleic acids, Tthe ultrasonicator (M220 Focused-ultrasonicator) enables accurate preparation of nucleic acid samples for many techniques of molecular characterization of nucleic acids, which enables the preparation of high-quality DNA libraries for high-throughput sequencing methods as well as methods for tracking epigenetic changes in DNA.


Automated microbial live-cell imaging and analysis system (oCelloScope)
Automated microbial live-cell imaging and analysis system (oCelloScope) that enables sensitive and detailed in vitro monitoring of biological growth and development in solutions (e.g. monitoring microorganisms in liquid culture). The system enables two types of analysis; optical density measurements and images acquisition. The measurements can be run through time for longer periods, and therefore, the system enables accurate growth measurements and analysis for low and high cell concentrations (range 103-107 particles/mL).The sistem integrates 5Mpx CCD camera with optical resolution of 1.3 µM. Detectable object size is in the range of 0,5 uM – 1 mm. FluidScopeTM scanning technology enables detailed 3D information acquisition at a single cell level. The cells can be monitored/measured without staining or other pre-treatment. The system is highly adaptable to various experiment layout, since it is compatible with standard microtiter plates, from 6 to 96 wells. Along with kinetic measurements the system can be used for analysing cell morphology on a single cell level as well as monitoring changes in cell morphology through time. The system is supported by dedicated powerful software (UniExplorer software) that runs the detection system and enables analyses of the acquired data. The instrument can be placed inside an Incubator or anaerobic chamber, if conditions beyond room temperature are required for the experiment. Real-time PCR instrument (ABI 7900HT Fast)
Real-time PCR instrument (ABI 7900HT Fast) is a high-throughput real-time PCR system that detects and quantitates nucleic acid sequences. Continuous wavelength detection from 500 nm to 660 nm allows the use of multiple fluorophores in a single reaction.

It has 384-well plate capability. Key applications include: measuring gene expression levels, detecting and quantifying pathogens, performing allelic discrimination assays (single nucleotide polymorphisms (SNP) genotyping). The system offers a choice of quantitation methods for gene expression. The absolute quantitation method determines target quantity directly from a standard curve. Relative quantitation eliminates the need for a standard curve by calculating quantitation values of an unknown sample relative to a calibration sample. The software completely automates data analysis, and can automatically set the optimal baseline for each well, as well as the optimal threshold for each primer and probe set (TaqMan Assays) or assay-specific primer set (SYBR Green Dye).

Real-time PCR instrument (Roche Light Cycler 480)
Real-time PCR instrument (Roche Light Cycler 480)is a fully integrated multiwell-plate based real-time PCR platform for highly accurate qualitative and quantitative detection of nucleic acids. It analyses true raw data, breaking free from the need to use passive reference dyes or normalization plates to get accurate results. It can be flexibly switched between 96- or 384-well plate formats and exchange thermal block cyclers in just a few minutes with no recalibration time. System employs a high-intensity Xenon lamp that emits light over a broad wavelength range (430–630 nm). The five excitation and six emission filters of the system can be used in any combination.

The system allows the use of a variety of fluorescent dyes and detection formats (monocolour, multicolour) for any current real-time PCR application. Key applications are: gene expression study (absolute and relative quantification), genotyping (SNP analysis) based on endpoint analysis or melting curves, High Resolution Melting which enables genomic researchers to analyse genetic variations (SNPs, mutations, methylations). The most important High Resolution Melting application is gene scanning - the search for the presence of unknown variations in PCR amplicons prior to or as an alternative to sequencing.

Real-time PCR instrument (ABI PRISM ViiA7 SDS) enables quick quantitative PCR, usage of 384-well plates, up to 6-plex analyses and melt-curve analyses.

Real-time PCR instrument (ABI QuantStudio7 Flex)
Real-time PCR instrument (ABI QuantStudio 7 Flex) enables quick quantitative PCRs, using a 384 or 96 tile plate, as the block can be easily replaced, a Melt-Curve Analysis and Multi-Target Analysis (up to six target sequences). The main applications are: determination of gene expression (absolute and relative quantification), genotyping (SNP analysis), which can be based on end-point analysis or the analysis of the melting curve.

Real-time PCR instrument (ABI QuantStudio7 Pro1)
Real-time PCR instrument (ABI QuantStudio 7 Pro) allows single or multi-color detection of nucleic acids (single or multiplex quantitative reactions). The system is very sensitive, allowing the detection of 1 copy of the target sequence in the reaction mixture. Linearity is ensured over 10 logarithmic orders. The instrument supports the performance of experiments with and without the use of internal passive control, such as ROX or Mustang. The instrument allows the use of different blocks, namely a standard block for 96 samples (10-100 µl reaction), a fast 96 block (5 - 30 µl reaction), a fast 384 block (5 - 20 µl reaction) and a block for microfluidic cards (1.5 µl reaction). No calibration (validation) of the block is required after the block has been replaced. The blocks enable fast heating / cooling (6.5 ° C / sec). The heated lid prevents the formation of temperature gradients in the vessels and prevents the concentration of the reaction mixture from changing during the PCR reaction itself. The 96 blocks allow rapid optimization of new protocols in one step with a very precise temperature gradient across the six columns across the block. Simultaneous implementation of six different PCR protocols in six columns, 16 samples in each column, is possible. The apparatus uses standard PCR plastic, such as individual tubes, comic tubes and microtiter plates in 96 or 384 sample format, as well as the use of ready-made microfluid cards.

Automated droplet Digital™ PCR System (Biorad QX200)
QX200™ Droplet Digital™ PCR System (Biorad QX200) consists of the automated QX200 droplet generator, the QX200 droplet reader, and the computer with software. The system enables preparation and analyses of nucleic acids in different samples with droplet digital PCR (ddPCR). The automated QX200 droplet generator provides fast analysis, reduces mistakes and increases reproducibility of the results. The QX200 droplet reader enables the use of TaqMan hydrolysis probe and intercalating dyes (eg. EvaGreen).

Automated Droplet Digital™ PCR System (Biorad QXone)
QXone™ Droplet Digital™ PCR System (Biorad QXone) is an automated system for preparation, execution and analyses of multi target digital droplet polymerase chain reaction, as it integrates a standard ddPCR workflow of droplet generation, thermal cycling, droplet reading, and analysis into a hands-free precision platform. The reader enables the use of TaqMan hydrolysis probe and intercalating dyes (eg. EvaGreen).The system enables absolute quantification of nucleic acids in multiple samples with droplet digital PCR (ddPCR) up to four or even eight targets simultaneously.

Digital PCR instrument (Stilla naica)
Digital PCR instrument (Stilla naica) by Stilla Technologies is a high-multiplexing digital PCR system. The semi-automated system comprises of Geode, which doubles as the droplet generator and PCR cycler, and Prism6, a reader with an integrated computer. Prism6 enables the use of hydrolysis probe in six channels, allowing for true multi-channel multiplexing, as well as the use of intercalating dye chemistry (e.g. EvaGreen). The system enables recovery of the sample following PCR and use in downstream analysis. Crystal Miner license-free software allows analysis in 1D, 2D and 3D, and provides visuals of the PCR reaction down to the level of individual droplets, including the raw images taken by the Prism6. Digital PCR instrument (Fluidigm BioMark HD)
Digital PCR instrument (Fluidigm BioMark HD) enables quantitave PCR and up to 5-plex analyses. The main advantage of the system is that it enables simultaneous analyses of up to 48 target sequences for 48 samples by automatic sample loading what increases the efficiency. The instrument enables also digital PCR for up to 36960 reactions simultaneously, or dPCR for 48 target sequences with 770 reactions simultaneously. It enables also absolute quantification of nucleic acids.

Robot for pipetting (Hamilton Microlab STARlet)
Robot for pipetting (Hamilton Microlab STARlet) enables automated pipetting of liquids. Instruments is based on the air displacement pipetting principle, comparable to the functioning of handheld pipettes. It has pipetting arm with four pipetting channels, which work simultaneously or individually. Conductive disposable tips are used to detect aspirated liquid and prevent contaminations between samples. Robot enables pipetting from 1 to 1000µl of liquid. It has a cooling system with cooling blocks for 2 ml tubes, 96 and 384-well plates. Sample dilutions for real time PCR reactions can be prepared and dispensed on plates using this robot. All parameters for pipetting (positions, amount, speed,…) are defined in the program. Software is very flexible with many programming possibilities.

Nucleic acid purification instrument (KingFisher Apex) - NEW
Nucleic acid purification instrument (KingFisher Apex) is an automated system for extraction of nucleic acids, proteins and cells, and is based on the magnetic separation of target molecules. With this system, target molecules can be extracted simultaneously from 96 or 24 samples, and chemicals from different manufacturers can be used. The system has a wide range of elution volumes and is therefore suitable for very demanding applications (whole genome sequencing) and more simple applications. The system also allows heating and cooling of the samples during the extraction process, which is especially important for more complex or sensitive samples. The isolation process itself is fast and takes up to 65 minutes. The system is equipped with a UV light, which allows decontamination of the system after each use and thus reduces the possibility of contamination of samples

Ultracentrifuge (Optima XPN-90)
Ultracentrifuge (Optima XPN-90) enables centrifugation at very high speeds, thus e.g. enables the sedimentation of viral particles or the separation of only these in different gradients. The ultracentrifuge enables centrifugation at a maximum speed of 90,000 rpm, has a built-in cooling and heating system, allows operation for up to 48 hours and operates in the range from 0°C to 40°C. Analitical balance and microbalance with a pipette calibration system (Mettler Toledo XPR56 and XPR205)
Analitical balance and microbalance with a pipette calibration system (Mettler Toledo XPR56 and XPR205) enable high-precision weighing and calibration of pipettes of larger and smaller volumes. The micro-analytical balance allows weighing with an accuracy of at least 0.001 mg with a repeatability of at least 0.0012 mg. The balances also enable weighing of samples, and with the installation of a moisture trap, it ensures greater accuracy especially when checking pipettes of smaller volumes. Chromatographic instrument (CytivaLifesciences Akta Pure 25M)
Chromatographic instrument (CytivaLifesciences Akta Pure 25M) is is part of the System for rapid preparation and concentration of biological samples with the possibility of biological analysis on the media. It is an ntuitive chromatographic system for the rapid purification of proteins, nucleic acids and viruses at the level of micrograms or tens of grams of the target product. The system supports a wide range of chromatographic techniques and meets the criteria that allow high purity of the product. Two system pumps based on technology developed for advanced systems. The robustness of the construction creates repeatable flows at low and high back pressures, which enables short separation times. The system pressure sensor is connected to the main pumps and continuously measures the system pressure and allows automatic flow adjustments to avoid the defined upper pressure limits. For the determination and separation of free nucleic acids, proteins and viral particles, the UV detector is designed to simultaneously detect multiple wavelengths in the UV spectrum and visible light spectrum from 190 to 700 nm (eg 260 nm for nucleic acids, 280 nm for proteins ). The UV detector allows simultaneous monitoring of three different wavelengths. The design of the flow cell together with the optical fiber technology allows a high signal-to-background ratio. The detector is automatically calibrated each time the device is switched on. The conductivity detector measures the conductivity of buffers and samples for online gradient monitoring. The integrated temperature sensor corrects changes in conductivity resulting from temperature changes. The conductivity detector has a wide measuring range and thus enables the monitoring of conductivity in various chromatographic techniques. Sample loading pump is designed to allow automatic application directly to the column. Using a sample loading pump saves time and is especially useful for working with large volumes. Samples can be loaded at a flow rate of up to 50 mL / min. During sample loading, the air sensor detects when the sample was finally loaded on the column and so we can proceed to the next step of the selected method without air entering the system or column. The proposed fraction collector provides flexibility, high performance and safety. The fraction collector is equipped with various cassettes that allow the collection of fractions in tubes or microtiter plates, which means that the samples can be in any form we need. The fraction collector is covered and prevents contamination of the samples with dust particles. The fraction collector has properties that reduce the chances of cross-contamination and spillage during the fraction collection itself. Software enables real-time chromatographic system control and accurate evaluation of obtained results.

Media preparation system (Integra Mediaclave 10 in Mediajet vario)
Media preparation system (Integra Mediaclave 10 in Mediajet vario) is part of the System for rapid preparation and concentration of biological samples with the possibility of biological analysis on the media. It enables rapid, efficient and standardized preparation of various culture media, which is the basis for working with all types of microorganisms. The system consists of several connected units, namely an autoclave and connected unit to it, which enables automated sterile dispensing of autoclaved media into petri dishes, a printer for imprinting petri dishes with different information and additional units for adding temperature-sensitive substances. The equipment allows complete control over the entire process in the possibility of adjusting the parameters to the needs of special media. Automation provides a continuous sterile environment and allows standardization of preparation and dispensing. In this way, a low measurement uncertainty of sensitive experimental procedures is ensured. At the same time, the system is universal and suitable for preparing media for a wide range of work. Quality assured prepared media represent the basis for work in all areas of work with microorganisms, both in the diagnostic and research field. Media preparation system thus represents a basic and key equipment in all research of microorganisms, such as study of antibiotic resistance, biological research of pathogens in other microorganisms, isolation of microorganisms from natural sources, detection of bacterial pathogens, development of use of bacteriophages for control of microorganisms, work with plant tissue cultures and verifying the effectiveness of inactivation of microorganisms.

Benchtop mass spectrometer for automated microbial identification system (MALDI Biotyper® sirious RUO, BRUKER) is a microbial identification system based on MALDI-TOF mass spectrometry (Matrix-Assisted Laser Desorption/Ionization Time-Of-Flight Mass Spectrometry). The system includes gridless microSCOUT™ MALDI ion source with extended pulsed ion extraction PAN™ mass range focusing; integrated source cleaning without the need of venting the system; 200 Hz smartbeam laser with minimum 500 million laser shots; high capacity vacuum system TOF analyser for linear-only measurements with positive and negative ion mode. The technology enables unbiased identification of microorganisms within a few minutes down to the species level. The system measures highly abundant proteins that are found in all microorganisms. It determines the unique proteomic fingerprint of an organism, and matches characteristic patterns with an extensive reference library to determine the organism’s identity. The system is supported by dedicated microbiology software automates the process of acquiring the mass spectrum and performing the match against the extensive reference library. System for identification of microorganisms using fatty acid methyl esters analysis by Gas Chromatography
System for identification of microorganisms using fatty acid methyl esters analysis by Gas Chromatography (Sherlock Microbial Identification System) consists of several interconnected parts. Since the analysis itself operates on the principle of gas chromatography, the system comprises a gas chromatograph, an origin of carrier gas, a column injector for sample introduction and corresponding detectors and recorders for data capturing. The system can also automatically inject samples for analysis, thus enabling a rapid and automated analysis of large numbers of samples. The data obtained by gas chromatograph is computer-processed and the profile of fatty acid methyl esters of the examined organism is compared with extensive data libraries that are included in the software. Thus, using these different data libraries very different micro-organisms, environmentally and clinically relevant aerobes, anaerobes, yeasts, and also the most important types of bioterrorist agents could be identified. The software also allows different approaches to data processing: plot dendrogram, 2-D graphs and histograms, and PCA analysis. It is also able to export data to other programs for data analysis. Growth chambers for plant and tissue culture breeding (PSI)
Growth chambers for plant and tissue culture breeding (PSI) enable breeding of plants under controlled temperature, light and humidity conditions. Two growth chambers with 42 and 50 shelves each are designed for breeding plants in soil and two growth chambers with 40 and 56 shelves each are designed for breeding plant tissue cultures. In the growth chamber designed for breeding plants in soil the temperature is set to 20°C in dark and 22°C in light, illumination of 70-90 μMm-2s-1, photoperiod of 16 h of light and 8 h of dark, and relative humidity of 70 %. Electric LED lamps and ambient lighting is installed. In the growth chambers designed for breeding plant tissue cultures the temperature is set to 17°C in dark and 19°C in light, illumination of 70-90 μMm-2s-1 and photoperiod of 16 h of light and 8 h of dark. Electric LED lamps are installed.


Plant growth chambers for separate breeding (Kambic)
Plant growth chambers for separate breeding (Kambič) consist of 4 separate self-standing chambers that provide for breeding of plants in soil under controlled temperature, light and humidity conditions. Outer dimensions of each chamber are 850 mm x 2800 mm x 930 mm (W x H x D) and inner dimensions amount to 750 mm x 1840 mm x 760 mm (W x H x D), which enables breeding of plants on 4 shelves.

The temperature can be set between 5°C and 60°C and is stable < ± 0.3 °C, which is rendered possible by a water cooled compressor generating set. Relative air humidity can be set between 30 % and 90 % and is ±10 % stable, which is enabled by an ultrasound humidifier and dehumidification by a cooling spiral. Illumination is provided by electric lamps Fluora, 6 x 20 W per shelf. Air circulation is forced by an electric fan with mild air flow. Plant growth chambers for separate breeding are especially suitable for breeding plants in quarantine conditions. Besides, they allow breeding in 4 separate compartments, between which there is no air exchange, which is very important in studies of a plant’s response to pathogen infections.

The temperature in the chambers can be set between 15°C and 35°C and is stable < ± 0.5 °C. Relative air humidity can be set between 40 % and 80 % and is ±5 % stable. Illumination is provided by multi-color LED lamps for every shelf. Plant growth chambers for separate breeding are especially suitable for breeding plants in quarantine conditions. Besides, they allow breeding in 7 separate compartments, between which there is no air exchange, which is very important in studies of a plant’s response to pathogen infections.

The large infrastructural equipment of IC Planta enables a wide range of research and applications in three fields:
  • Microscopy (Transmission electron microscopy and Confocal fluorescent microscopy),
  • Modern techniques of molecular biology by using five real-time PCR instruments, four digital PCR instruments with the belonging equipment and robot for pipetting that offer all currently available different PCR technologies (including droplet digital PCR) for quantitative analyses of target DNA molecules and gene expression analyses in different biological samples,
  • Breeding of plants and tissue cultures in controlled quarantine conditions by a set of growth chambers and quarantine greenhouses with the belonging equipment.

 



IC Planta’s equipment supports research activities, bodies of ministries, enterprises and educational activities. IC Planta ensures collaboration between researchers of different research programs, projects and institutions. It facilitates connections of researchers with the users of this research, which are other budget users and various industries, as well as facilitating connections between research activities and educational processes.

The number of users of IC Planta’s infrastructural equipment per year is between 60 and 80. The subjects of research and analyses carried out by the Centre Planta’s equipment, are extremely diverse.

IC Planta’s infrastructural equipment is used for the research activity of different research groups from different ROs. It is used to carry out research programs financed by the Slovenian Research Agency, for the performance of research projects financed by the Slovenian Research Agency, for the training of young researchers, and for international research projects, among them EU framework projects and bilateral projects.

IC Planta’s infrastructural equipment is used for applied projects ordered by different Ministries and agencies (Ministry of Agriculture, Forestry and Food, Ministry of the Environment and Spatial Planning, Ministry of Defence and Slovenian Research Agency). Besides, IC Planta’s infrastructural equipment is used for projects that serve to support different public and governmental bodies and the performance of public service. In the frame of the annual Expert projects in the field of plant health protection, the equipment was especially intensively used for the support of the activities of the Phytosanitary Administration of the Republic of Slovenia and the Inspectorate of the Republic of Slovenia for Agriculture, Forestry and Food. IC Planta’s infrastructural equipment was also used to support the Ministry of the Environment and Spatial Planning, Ministry of Agriculture, Forestry and Food, Ministry of Health, and bodies of inspectors in the field of genetically modified organisms.

IC Planta’s infrastructural equipment is used for applied projects which are ordered by commercial enterprises, which directly supports the industry, environmental protection and agriculture with the development of new technologies and performance of highly specialized analyses on the basis of good laboratory practice. IC Planta’s equipment supports technological progress in the fields of plant, animal, pharmaceutical, medical microbial and food biotechnology, and development of methods for specialized analyses in diagnostics of plant pathogenic bacteria, viruses and phytoplasmas and genetically modified plants and plant products.

IC Planta’s infrastructural equipment is used to support the performance of different subjects at different universities.

Research and analyses, where IC Planta’s infrastructural equipment is used, especially the equipment connected with GMOs, importantly contributes to the increase in the quality of life, to awareness of various environmental issues and because of its wide response also to the creation of public opinion.
