Setting methodologies for studies of potato immunity at single cell level

Within this project we will establish methodological basis for high-spatiotermporal resolution monitoring of potato responses in interaction with different microbes. We will develop approaches at the level of single cells and and the tissue level. For the analysis of individual cells we will use single-cell sequencing approaches following transcriptional and chromatin rearrangements. Additionally, we will follow translational activity in infected cells. For analysis of responses on the tissue level we will test applicability of genetically encoded luciferase sensor plants. If this approach will not be succesful we will employ tagged microbes in combination with microtissue dissection to get spatial resolution of responses. As much as time will permit we will gather first datasets for each of the developed methodologies. This will altogether provide additional preliminary data and proof-of-concept evidence that were requested by the reviewers of the GUIDECROP ERC project.
